I know he used haki, but he was still caught off guard because he expects tashigi to be weak and use a sword, but she attacked him while he had his guard down, and she was using smokers kairoseki weapon, which he didn't expect, you can clearly see he was confused which is why he got tagged and nulled.Off gaurd when luffy has observations haki and can fight blindly

Doflamingo one-shot Smoker whilst at full vitality. He was in a very weakened state when Gear 4th "bodied" him (He actually stalemated him for an extended period, btw). Just sayin.
