First Round: Zeref is quite advantaged there honestly except for genjustu not sure what can Madara do. So Zeref via durabilty negation,regen,time stop (thou he never used this in battle so I am not sure).

Second round: hmm... I am gonna wait to see more from Zeref before saying anything else, but for now Madara has this thx to rinnegan hax.

Round 1: I dunno. Madara could put him under genjutsu and take his time dicing him up while Zeref can use his death magic on him.

Round 2: Zeref can't kill Madara but Madara can do so with his Soul Manipulation or the King of Hell (I think).

I'll think about it more later but if those don't work on Zeref, it's likely to be inconclusive.

Okay, so:

Round 1: Since they're in character, Madara wouldn't use genjutsu and Zeref wouldn't use his death magic. But Zeref has stuff like Law while Madara just has precognition, Fire Release and brute force with Susanoo. Zeref should be able to take him out with Law but Madara would in turn use Izanagi and knock him out from behind or make sure to place him under genjutsu and do the same. I go with Madara for now.

Round 2: Zeref can do nothing to Madara but Madara has Soul Manipulation and One Hit Kill with the King of Hell, can suppress Zeref's magic with Wood Release or knock him out with the pollen from Wood Release. I go with Madara here too for now.

R1 in character madara wouldn't use genjutsu and zeref can't contro his power and zeref has many more hax like law,exchange,time stop,demon and all ability from book of zeref

R2 zeref can kill him with death magic since that can also kill immortal,demon,law,time stop.... Like R1 and non of zeref demon can kill him(soul,poison,etc) except e.n.d(posibly more)

I give zeref for both round

Sage Mode Madara is City level and a glass canon. It's just like Madara can't kill Zeref in Round 1 too so I don't see the problem.

Oh, I was looking at his normal durability. Regardless, the situation for Zeref in Round 2 is the situation for Madara in Round 1. Zeref has too much hax for Madara and the only way I see him winning is with Izanagi + a surprise attack like I said above.
