How fast can she activate her stuff?

Before people with speed comparable to her own can react in the case of the soul mirror transformation thing. Most of her attacking moves require her to throw Tarot cards and/or summon mirrors before the attack is used. Even if TWOH was turned into a mirror DIO might still be able to fight back with his vampire powers.

He spun three times and pointed into the air before using the ability, after that point it was instant. But due to Zonda's personality and Gunvolt and Copen being able to do jack about it (Legs frozen in ice and all) it could be either way. In the cutscene it takes a little less than two seconds for the gesture Zonda makes to end. Though it should be noted this was base Zonda, Reverie Zonda might be able to do it faster.

Summoning stands is at least fast as thinking, so DIO gets my vote.

Yeah. It's possible Zonda's ability is thought based and Zonda posing was due to her personality, but Possible thought based < Known thought based. There's also the fact that DIO would still have a much weaker TWOH (Unless stand damage reflection would cause him to shatter too) and even if he did lose it entirely he still has his vampire powers to at least fight back.
