Unlike Ragyo and Ryuko, who are humans infused with life-fibers, Nui is completely made of life fibers, so her heart cannot be crushed and has to be cut from both sides simultaneously otherwise it would regenerate. Simply piercing it or cutting it open wouldn't kill her.

But she can be defeated, there's just a specific you have to kill her otherwise she won't die. She literally can't die unless she is cut from both sides at the same time, that's how her Regenerationn works. She was even stabbed through the heart by Ryuko, pretended to be in pain, then pulled it out to show her while laughing about it. Heck, Ragyo survived having her head cut off by Satsuki, but because it was only cut from one side she was able to reattach it, and her Regenerationn is about equal to, if not lower than Nui's.

Doflamingo should win this battle at least 7 out of 10 times. Nui's physical feats and stamina are outmatched by Doflamingo's, their defenses are equal, their speeds are pretty close in comparison, both have the ability to control the other, though those powers can be easily stopped by both parties. Nui has superior Regenerationn but Doflamingo has much more versatility. They are both tactical geniouses, but Nui has shown to be MORE arrogant than Doflamingo and dropped her guard relatively easily in two fights, resulting in losing her eye and then her arms towards the end of the series. Doflamingo's clone--via feats--has shown to be stronger than Nui's version.
Doflamingo would most likely have to use his awakening, but once that happens, he wins via Everwhite and God's Execution fairly easily.
