Destined One's suit gives Kamala extra appendages and piercing damage, that'll help to counter alotta Luffy's stuff,
Add to that good rate of regen and versatile experience, alongside more variety such as dodging via size manip, Kamala should take this high-diff.

Luffy holds a considerable AP advantage here; scaling a good deal above 8.52 Tons while even casual. If the fight actually goes on long enough Luffy should be able to improve due to Accelerated development. If required Luffy could keep Kamala at bay similar to what he did with buggy by using his superior lifting strength to completely overpower her. Assuming the fight drags on Luffy should still take this due to superior stamina.
Also does Kamala have a way to overcome Luffy's blunt force resistance (Nevermind for this last point.).

And Luffy at his highest scales pretty high above that, but you'd have to ask Cal about that.
