Okay, i belive that this two characters deserve some attention, they come from the beginnings of the indie games which caused a great impact on the videogame industry.

9-A V/S 9-A: .

Round 1: Speed equal, both fighters start 50 meters from each other, they are in-chacater.

Round 2: Speed unequal, both fighters start 100 meters from each other, they are bloodlusted and also they had five days of prep-time.

Round 1: Quote easily thanks to versatility and equipment

Round 2: Asuming the right collection of items (i.e. Kamikaze + Pyromania, Gnawed Leaf + any meatboy or daddy longlegs, Sharp Plug + Blank Card + XIX Sun), Isaac will either heal faster than Quote can damage him, or be literally unkillable to begin with, while also being able to do serious damage to Quote, so Isaac takes this.
 
Quote takes round one for having the best equipment at base, aswell as skill and experience, round two I'm going whit Isaac, since while speed would give quote a brutal advantage, many items (as far as my memory goes) from Isaac will help negate the damage made by Quote, aswell as tired him, until Isaac has the opportunity tho take him down.
 
Round 1: Quote for reasons above

Round 2: Daddy Longlegs + Gnawed Leaf or any other broken combo. In five days Isaac could have likely collected every item
