She used it on Aggregor immediately even though she didn't know how powerful he really was after absorbing it on 5 Aliens.

Squid's playing has npi so Squid can destroy her spirit with his Clarinet playing and prevent her from regening.

I don't see Ghost exploding or dying, and she has "Low-Godly" so she would be able to regenerate herself as it's not mentioned that Squidward has Soul Destruction and Regeneration Negation (Low-Godly) in her profile.
