If they aren't even aware they are in a fight? I don't think Puff would attack first, and the Enderman would most likely get a first hit in. Considering Puff's higher durability, he would most likely survive the first onslaught and then proceed to time stop and just finish up the Enderman.

Puff wouldn't fight back, at all. Iirc, he has actually ran from combat before.

I'm just going to disregard speed when putting my thoughts on this fight, as neither have shown any outstanding feats of such.

First and foremost, Puff holds the greater arsenal of abilities at his disposal via his reality warping, but the Enderman's teleportation is gonna be problematic. The Enderman's habit of teleporting behind the player when being attacked could prove quite useful here against Puff, since the Enderman could thus close the distance between himself and Puff and engage him in melee combat, where the Enderman would hold the advantage.

When it comes to durability, both are hardy beings, but I'd say Puff might be more durable due to surviving being crushed beneath many rocks quite easily: https://youtu.be/N_5Z7UhVjGA?t=20m29s so he could survive many of the Enderman's blows.

When it comes to smarts, there's no debating it, Puff is WAY smarter than the Enderman. He might learn to anticipate the Enderman's teleportation and prepare himself for a possible sneak attack. He'll also do his best to avoid getting into close range of the Enderman and use his reality warping to keep the Enderman away.

So overall, this would be a long fight, but I'm gonna give it to Puff for a combination of his durability and reality warping, though the Enderman's teleportation and superior melee skills are gonna make things hard for Puff.
