Power difference between tier low 2-C and tier 2-C?

Is there an actually quantifiable difference between the power to destroy one 4D space-time continuum and the power required to bust two 4D space-time continuums? Like would being 2X Universe+ make you multi-universal?
 
I am uncertain. The universes likely do not share the same spacetime continuum after all. We have tended to go by demonstrations or stated definitions regarding this.

It can vary, though if you are shown to be significantly superior to a Universal+ character, it is usually grounds for Multi-Universe level. Getting direct statements, such as a character being hundreds of thousands or infinitely superior to a universal+ character give us more to work with, but as Ant said, it is a case by case thing.
