In character, no prior knowledge or prep time, victory by KO (since this is a boxing match), Takes place in the Smash Bros. Boxing Ring.

Yup, in the 1987 version he defeated Super Macho Man.

In the 2009 version, he already has more abilities to contribute to this so it's safe to say that he takes all 3.

Super Macho Man is able to defeat Don Flamecon with no effort, the same Don Flamenco who knocked a charging Bull over the horizon with no effort.

Because Little Mac > Super Macho Man > Don Flamenco seeing as Super Macho Man is the strongest character in the 1987 version of Punch Out, and the second strongest ( behind Mr. Sandman) in the 2009 version.
 
But the bull-punching feat didn't happen until the 2009 version, and Mr. Sandman was in the '87 version, and he one-shot everyone Mac had ever fought in that game up to that point, so if we're using feats that didn't exist yet, wouldn't Sandman be the strongest?

Yeah but the 2009 version is a remake, the actual sequel to that is the Snes version. Though Mr.Sandman replaced Mike Tyson in the remake, so I guess give it or take you can either put him in the list or not.
