Tsunade wins handily. First and foremost, she has a hefty AP advantage here. Secondly she's way more skilled and versatile. She has better Regenerationn, durability negating attacks as she can do internal damage, and some more abilities.

The only real advantage Hancock has is flight, but he doesn't seem to have any ranged attacks to take advantage of it, so it's a moot point. Not that he can hurt her anyway.

This seems like a stomp in my opinion.
