Let's say a verse has a 1-B Cosmology, and a character manipulates the plot. Does this mean they have 1-B Plot manip?

If the plot expands through all of it and they are shown to manipulate the whole plot, yes.

So long as they're referring to the actual plot, yes.
It should be noted that this is simply the perspective from which they manipulate the plot, this doesn't mean they have 1-B range or capabilities.
