Simba vs Shere Khan

Hmm...
If we want to go with the original, written Jungle Book canon, then Simba would likely win. The conflict brought by Shere Khan was due to the tiger being a cripple. He was forced to hunt humans instead of sticking to the wild, which the other animals were afraid would cause the humans to retaliate by setting fire to the entire jungle. He would be at a disadvantage against the younger, healthier Simba.
But I'm pretty sure you were thinking of a strictly Disney duel. In that case, I'm a little torn. I would like Simba to win, but I think Khan has an advantage. This is because Simba is heroic, Khan is a vicious fighter, and that these two traits would likely skew the fight in favor of the tiger, especially because that's in-character for both of them in real life. It's still a "Pirates vs. Ninjas" debate, but it's rather interesting.
I'll give it to Khan on account of being a dirty heel and Simba an honest brawler.

Simba defeated Scar, a vicious fighter. And he managed to repel all of the Zira's pack alone as well (Even more, he threw away various lions with a punch: https://youtu.be/gdH_WclU42Q?t=110)

Scar was not so much vicious as he is a pragmatist (throwing dust in the eyes, pleading for mercy, etc.). He was throwing dirty moves left and right, which in my eyes is harder than someone who is just outright vicious.

Welp. From The Wandering Shepherd's video (1:51 onwards), Simba wins via enough strength to throw around lions roughly his own weight if slightly less with paw flicks.
