Niko would wreck him with pressure points and krav maga, but through sheer speed and acrobatics, Leon gets a huge advantage.

Leon has just as much if not more experience than Niko in martial arts and combat, honestly.
Niko's limited experience and training in the yugoslavian wars years before the events of GTA IV hardly stands up against the prolonged military training Leon has had as a policemen first and has a secret government agent.

And while Niko has gone through a good load of crap and difficult situations, Leon has dealt with all sorts of giant monsters, undead folks and animals, superhuman people and so on, a whole other class compared to what Niko has accomplished.

As previously said, Leon is also more agile and I add he is likely to have a better equipment and indeed more experience in using it in a variety of situations.

Without a doubt, to me, Leon takes it.
