Hmm this is a tough one. I'd be willing to bet John is superior physically and in hand to hand combat considering how much of a "peak human" he was hyped up to be and the fact that he was fully confident in being able to take on Sam without the shield before the Dora Milaje stepped in and interrupted.

However, Sam has his wings here and is probably smart enough to know that taking on John hand to hand might not be the best idea so he could easily take to the skies immediately putting John at a disadvantage. John still has shield throwing as an option, but John pretty much throws the shield the exact same way Steve does (albeit with a difference in throwing strength), and Sam's been around Steve long enough to know how that works so I'm taking my bet that he'd be capable of outmaneuvering John. He can also dispatch Redwing, making things even harder since Walker's dealing with 2 flying enemies that both have weaponry capable of killing him.

I'll vote Sam with mid to high difficulty for now.

And I'd argue he wins easily, too. John is just naturally arrogant, so I wouldn't take his confidence in taking Sam on too seriously tbh. Sam has much better skill feats, and his range advantage is just too much.
The fact that he was able to somewhat contend with Serum John says a lot.
