I don't think Luffy can kill Makima here. He doesn't have a way around her ressurection. Though, Makima doesn't have many win conditions either.
Looking at his profile, Luffy doesn't seem to resist the Mold Devil's abilities, nor can he escape from Hell if he's sent there by the Hell Devil or the Spider Devil (though, actually sending Luffy to Hell will be pretty difficult with his various speed amps and precognition).

He doesn't need to. He needs to just think and use the ACoC he spams the **** out of and that automatically knocks her out as many times as it takes to drop her.
Don't throw non-busted characters at Hakigods, or you will be hakidicked.

The damage she'll take from being knocked out will just be transferred to someone else.

It's not damage. She just drops because her willpower isn't good enough to resist getting literally dominated.
She doesn't have supernatural willpower nor a resistance that matches up with Conq Haki, let alone in the layers required.

It's not described anywhere, she simply doesn't react, and remember the scene where she confronts the Darkness Devil, she also doesn't react to the pain of having her hand crushed.

IC, Makima doesn't really attack and just relies on her passive damage transferal so she won't really do anything. She's just cocky. Nothing is really stopping Luffy from just ACoCing her and just GGs.

Law reversed a genderswap via Haki, among other bio things from Devil Fruits.
Which isn't Lethal and is a LOL worthy amount of pain for someone with as much pain tolerance as Luffy.
