Is it purely destructive power? Is it some kind of averaging of all their stats? What if someone has a durability that massively outstrips their AP?

What determines Tier?

Tier comes from Attack Potency, which is the raw destructive power that a character/weapon/etc can produce

In addition, Attack Potency/Lifting Strength/Striking Strength may not be directly correlated, and characters may be a certain tier for only one attack or ability.
Nevertheless, in most cases, they tend to be at least somewhat related, but there are quite a few exceptions.

A characters tier is based on how much damage it can do to either an opponent or like you said the destructive capability they hold.
For example if character A can hurt character B, and character B can tank attacks that can destroy islands like nothing, than by default character A has island level AP, which puts them as tier 6-C.
Now if a characters Durabilty like you stated is drastically larger than their AP, they are sure still ranked as whatever tier they can dish out, however we do note that their Durabilty is at this tier.

I forgot to mention , your tier can also have something to do with hax as well, you can physically have way less DC, but still be ranked like star level spending on your hax.
Like if character A has Wall level strength, but can summon stars and throw them, than he would be ranked tier 4-C.
