I'd say Tytos, with plenty of difficulty, altough Spindle could win just because of his weapons. (Btw, Spindle doesn't uses knives... )

Tytos is physically the strongest there, even without the X Drug. A. R. Tombstone should be more skilled than Spindle, but Spindle has feats such as tanking point-blank explosions and Molotov Cocktails. Also, both Spindle and Tytos have guns. So, all in all, I think that

(Physically talking) Tytos > Spindle >>> Tombstone.

(Skill) Tombstone/Tytos >>>> Spindle.

(Weapons) Spindle >/= Tytos >> Tombstone.


Seeing like that, I think that Tytos should win with a lot of difficulty.
