Earth's Guidance hard counters the Cobra's stealth and camouflage, and Hitokage would make quick work of the Cobra's scales seeing as it can melt stone and its entirely in character for Snake to coat the entire area in fire, and snake has a frankly massive target meaning its unlikely that he would miss

I thought that size and poison would help Giant Snake but seems like not....

Size is kind of a detriment against someone like Snake whose weapon relies on spreading as much AoE damage as possible

Poison only works if the Cobra bites him. It would kill if the Cobra does bite him but snake's fire is much more likely to kill first.
