Unsure if Lightning can beat that regen.

Lightning's Lifting strength and stamina are on another level compared to Ryuko's. Taking into account Ryuko's better Regenerationn, the battle would probably get dragged out. However that might be bad for Ryuko as her stamina is "Very High" compared to Lightning's "Nigh Limitless"
Lightning also has alot more battle experience, and is more level headed than Ryuko.

Lifting Strength and stamina won't really help when Ryuko will just keep getting exponentially stronger every second she's in combat due to her passively absorbing life fibers to increase her strength as well as her getting stronger from any damage that lightning manages to deal to her via her zenkai

I still say Lightning, she has Soul and Time Manipulation to draw on, which Ryuko has no defense for, besides absorbing Life-Fibers to get faster over time.
We also see Lightning pull a soul out of a guard in the opening cinematic of LR, even if this doesn't kill the guard, in game she absorbs souls to empower herself further. The more powerful the soul the bigger the boost, and with reality on the verge of collapse, that is a lot of souls.
Lightning in LR is ready to kill enemies, human or otherwise, but isn't in a bloodlusted state; so her killing Ryuko isn't much of a stretch

Ryuko also gets faster by regenerating from damage, and the only time manip Lightning has that is actually going to help her here is Overclock and that only lasts for a few seconds. Plus from what several people knowledgeable on Lightning have said that she doesnt lead with soul manip. By the time she thinks to use soul manip Ryuko will have both adapted and absorbed enough life fibers to be fast enough to blitz.
Also, i find it funny how Lightning's stamina is limitless for fighting for 13 days when Ryuko and people with stamina comparable to Ryuko can go for a month without rest while also sustaining normally life threatening injuries.

for reference for Ryuko's stamina, she's able to wear a Kamui, which is 100% Life Fibers, without any ill effects, when an ordinary person wearing clothing made of only 50% Life Fibers gets overwhelmed to the point of becoming a crazed monster. She withstood a day-long beating in her base form from Mako and was no worse for wear, and fought all four of the Elite Four back to back without a break despite taking severe beatings in each fight. Without Senketsu she crossed Japan in a day to collect all of his pieces, taking out all of Honnōji Academy's students in the process, and later took down an entire army of COVERS singlehandedly without Senketsu. She's able to shrug off injuries such as being bisected, being impaled, being torn to shreds, having her heart ripped out, losing several gallons of blood, and skinning herself alive [a feat which was marked as something that should have killed her as she snapped her own Life Fibers] and shrugged off being impaled hundreds of times in her fight with Ragyo despite going through a protracted battle before it. Her stamina is also comparable to Satsuki' who was able to fight no worse for wear after being subjected to over a month of non-stop torture and immediately after escaping torture went on to fight off a small army of COVERs using nothing but hr toenails as she didnt have Junketsu and her hands were bound. She's also comparable to Nui, who was able to work nonstop on creating Shinra Koketsu even after her arms were cut off.
