I wonder how do you draw lines on scan? Is it nessesary to measure observational error?

Just use paint.net or GIMP to draw lines, they both tell you the appropriate pixel-line count. You need to make sure that the "measuring stick" you're using to get the size of the other object via pixel-scaling is close enough to the "measuring stick".
