What is fushi gonna do about Alucard's regen and casull??

Potentially burn him or, if that doesn't work, fight him until the souls are drained up and then kill with silver bullets or something.

Fushi needs to do a stake through the heart thing or use holy weaponry to take 1 soul from alucard. I don't think fushi knows that.

He does indeed not know that. Unless maybe he uses the form that can see and talk to spirits. Then he could maybe figure out.
However, destroying Alucard to the point of destroying his heart is pretty basic stuff. And Fushi does have a habit of using roots to impale people at that.
What holy weaponry is concerned, he might get the idea to copy Alucard's guns / bullets at some point just because he will learn during the fight that those are some pretty strong weapons.
If Alucard decides to go Level 0 Fushi might also end up copying undead and learn the effectiveness of them on his own body.
