Winchesters have the Colt with 4 bullets.
Winchesters have very limited prior knowledge (Ash could only find out that one Survivor said "believing is the key")
IT has already made several victims, in 3 days he will kill 2 more and then rest, that's the time they have to find his lair and kill IT.

Although this seems like an interesting confrontation, the truth is that I don't know if the Revolver is capable of negating the Low-God regeneration. The revolver is capable of killing the Demons that regenerate from being reduced to ashes

It was accepted as negating Immortality without specifying the type, tho
Also, from believing The Colt will kill him forever...he dies forever whenever they use it, no?

I think that should be updated, but if they believe it could kill him, it might work. After all, there would also be the option of incapacitation

Well, now that it has been established that at least Sam and Dean have a chance to kill him, the next thing to find out is whether they discover him and... They have every chance of succeeding. I don't know how this version of IT works very well, but it seems that his arrogance and the fact that he already has a recent history make him quite easy to defeat, since Sam and Dean have been able to handle cases in a matter of weeks or within a few days and even close them in hours if the enemies confront them directly. It would all depend on whether IT tries to attack first or not
 
Think they would be searching throughout the city and eventually find his traces
They would be able to find the sewers/the house with the well quicker than the kids, but they would be needing to be together to pull each other out of the illusions (love takes people out of them). They also have to discover how powerful is believing against IT, to get any condition against him

Separate them? Gg.
Not believing enough before being able to use the Colt against IT? Gg.
Hence why I think this match is pretty fair

If IT were to discover that it is being hunted and is preparing to attack first, it would likely resort to using its mental and manipulative tactics, probably trying to play on the weaknesses of the Winchester brothers. However... ironically, the weakness of both at the hands of IT would actually be their strength. The willpower that both develop when they are in danger or when their loved ones are affected is simply brutal; they reach an incredible tenacity, likely enough for several of the mental tricks to not work. I would even dare to say that it would allow the brothers to understand IT more quickly and defeat it in a direct confrontation, as Dean is quite pragmatic and tenacious while Sam is more intellectual and cunning, making it likely that they would figure it out in any case

I don't know if IT has any way to attack to kill directly without being swayed by mental tricks or its arrogance, so I think Sam and Dean could take this
