Well, the chemical energy can be calculated, but the volume, mass, or density is hard to swallow. And if something is described as "An Antimatter bomb" we avoid assumptions that the whole thing is made of anti-matter.

Question is vague, but usually yes. E=mc^2, use the mass of the antimatter plus the matter it destroys as m, and the speed of light as c. This will give you energy in joules.

Though even so, as I mentioned above, the density of Antimatter isn't always something that can be properly calculated. Though if there's confirmed mass; then yeah it's that formula above. And when something is referred to as "Anti-Matter Weapon" it's not always assumed the entire mass of it or its projectiles are 100% antimatter.

If it's not stated, you can usually rely on the destruction it causes, or worst-case-scenario, assume it's similar to an equivilant real bomb, and use the density of that bomb's volatile material for reference.

I agree on just using destruction values if there's no proof on amount of Anti-Matter
