Yes and no. He needs to channel the natural energy of the horse, he doesnt NEED to ride it, but he does need it.
For example against Funny, he couldn't ride it, given his horse was dying, but he used a steel ball to have his horse kick him, which was enough to qualify for the natural energy and thus activated Act 4. Once activated, Act 4 and the Infinite Rotation lasts awhile even afterward, at least a few minutes, probably a tad longer.
There is one time, at the very end of the Part (like the last few panels), where Johnny manifested Act 4 without a horse, but we don't know how he did it, just that he did (Maybe it developed kinda like Heaven's Door?) or maybe he used the steel ball memento he had from Gyro to activate it, but due to being vague I wouldn't use that even though he technically can do it with a horse via unknown means.
But yes, horse is needed like 90% of the time, or something similar to use the natural energy, but Johnny doesn't have anything like that besides his horse.

Even Act2/3 has some hax via the spatial holes that negate durability.
Though Johnny is a huge glass cannon even if his bullets are kinda ridiculous.
