That doesn't really matter when Wesker can't even hurt Homelander due to the stat gap.
All Homelander is going to do is chase Wesker with his heat vision and hopefully try to close the distance to hit him. Wesker is much more skilled and isn't going to be overwhelmed by a 3.6x AP difference.

You're missing the point. Homelander no-sold the calc he scales to, so Wesker being several times weaker than that means he can't do shit to Homelander.

This is Dynamite Entertainment's Homelander, which is only 14.8 tons (8-B) in scale. How much would you scale Homelander to make the difference greater than 3.6x?

Oh, I thought this was the show one who no-sold that factory exploding with him at the dead center unscathed.
 
Ok so the only thing going for Homelander is that he’s greater physically, better AP and LS, and heat vision. Now this would be enough to actually take Wesker down, however Wesker is far more skilled and experienced in combat and dealing with things just as strong or stronger than him. Couple with the fact that Wesker has far higher reactions, Homelander isn’t tagging Wesker (even with speed equalised). I guess Homelander’s scream rupturing Wesker’s eardrum could catch him off-guard but Wesker has better regen and resisted far worse pain so… I’m voting Wesker, unless Homelander grabbing Wesker and lasering his head off is the more probable outcome than Wesker beating Homelander to death.

I can see the heat vision hitting Wesker. But the issue is that it's heat vision. No mass. So that doesn't stop Wesker from closing distance where not only he is most comfortable but where heat vision is less effective. And I don't see Homelander landing the HV reliably enough to put Wesker down before distance is closed. Besides, Wesker has regen.

Homelander's screaming will have minimal effect. It will hurt but it wont' cause actual harm and Wesker has pain resistance.

Actually, Comic Homelander also no-sold his value. Modern Supes like Maeve are far superior to Supes who can melt tanks and she couldn't deal any damage to him.
 
The 8-B Homelander scales way above the linked value. The feat of melting tanks is by Crimson Countess, who is far weaker than Maeve, who Homelander tanks hits from with no harm and then one shots.

So I'm not sure Wesker can even damage him here.

Actually yeah, Maeve (who is vaguely weaker than Homelander but still superior to Crimson Countess) only left a scar where it took Black Noir (someone vaguely stronger than Homelander) to actually maul him. Yeah Wesker isn’t going hurt Homelander, if he does it would be after a really long time of hitting and shooting at the same spot. But in this case Homelander could one shot Wesker if given the chance.

This seems like a stomp, sadly. Give Wesker some more stuff like Uroboros and prior knowledge and things might change.
I still don't understand how One-Shot works. Are we supposed to assume that the opponent will automatically increase to 8X (7.5X) just because he did in his verse, or would he only increase it near that point?

You can't really quantify one shot differences in verse (unless they're... well, quantified in the verse) but Homelander scaling two one shots above a value that is already 3.6x Vesker's means Homie probably one shots him. Especially with hv since it scales above his physical durability, i believe.

Now Wesker has Uroboros
So he lost his higher reaction speed but now has the wincon of potentially infecting/absorbing Homelander. However, since John has prior knowledge and is bloodlusted, he'll just immediately use heat vision and keep going until he's sure Albert is dead.

Ok so Wesker has a viable win con now of poisoning Homelander. Also I forgot to mention because it didn’t cross my mind but since the setting is in the RPD there’s a bunch of zombies, Lickers, even Mr X and other tyrants (such as Nemesis) that Wesker can command via his psychic link with T-Virus B.O.W meaning that Wesker has cannon fodder to distract Homelander with. Which helps greatly since one of Homelander’s weakness’ is his attention span whenever he uses his enhanced senses.

Actually, ignore this part. El Gigante's feat is only 1 ton, so it's 4 times weaker than what Wesker is scaling to right now. He apparently scales to a whale monster that can also tank rockets, so no oneshot scaling.

No outside influence unless OP wants them to be there.

His enhanced senses aren't passive but need to be activated, and uh... Why exactly would he need to use them? Wesker is 6 meters in front of him.

I want to bring up that Wesker ALSO no-sells his current rating.
He's one of the few who can tank rocket launchers. Multiple of them. Even creatures like the Tyrants can survive getting hit by one, but go down with two.
You can hit Wesker with several RPGs in his boss fight and it only stuns him.
So, that probably means the difference between them is still close to 3.5x.
