Anyway, any Anna wins via Sleep staff. Kai is put to sleep.

But then Kai eventually wakes up, Sleep Staff isn't permanent and Anna has no way to kill him

Hexing Rod wouldn't fully null his regen tho, just up to a "halfway point", and Hexing Rod already deals the damage down to that halfway point

Kai's Regenerationn is manual yes, he needs to use OverHaul on himself to regenerate. However if his arms are destroyed he can't use his Quirk. So my question is what does Anna start out with in character?

That's confusing. I can't say for sure since i don't know what she'll use first. So inconclusive, Kai either destroys her on the molecular level or she haxes him to death.
